It depends on how it's done, for me. If you get the shade or style just slightly wrong, it looks less like lipstick and more like the girl's sporting a mustache (look at the Disney character Kim Possible for what I mean); I think that'd be a bigger turnoff for more people than a simple line for the mouth, but that's just me.

Are we talking about lipstick -strictly- found in animated series or in anime style artwork, too? Whether a female character has lipstick on or not in anime has never bothered me, nor have I ever found a character who looked silly with it drawn "in." I do think more mature, milf styled women in anime always look good (or better) with lipstick, especially if it's red.
In the case of this OVA, the lipstick looks fine imo, but what you should really pay attention to is the VN. The eroge this is based on features the artist "ogin bara," who quite honestly draws really nice lips. I don't think his characters would look as nice or unique without lipstick/defined lips.
You guys should really check him out btw, the girls he draws look like freakin' angels.
Oh, and the oppai. There's lots of grade-A oppai.
