After you beat the game, go all the way back to the room where you started, and activate the computer next to that red and blue door looking thing. Its sort of like a post-game area.

There's a whole extra area with new enemies and this boss. The first room of the game has a teleporter that activates after beating the rest. After going through, you can go right into the extra stage, or left to a gallery-type area where you can re-fight bosses and encounter all enemies, including the new ones after you beat the extra stage.

I'm in that extra area that you reach from the teleporter in the first room. I can go left, but the way right is blocked. What do I do about that?
I've already finished the game on hard mode even...

I don't know about that, maybe you have to find all the items or something. Unless somehow you're talking about the gates in the gallery that block off the extra area's enemies and boss.
