@anon2 it's from the title sequence of one of the OVA/ONA/Webisode/whatever you want to call them. I can't remember which off the top of my head but maybe 13 or 14?

Kirino laughing in the background just shows how much she "cares" for her nearly dead kind brother Kyousuke.

Manami is a bitch. She wanted Kyousuke to herself when they were younger, and so she took advantage of a situation to degrade him. Kyousuke used to be outgoing, and energetic. He was on the athletic team, he had the highest grades in elementary. One day a friend got hurt when the two were mountain climbing. The class blamed Kyousuke, so he went to Manami for advice, because up till then she had seemed like a good friend. She told him that the accident occured because he was normal and trying too hard to be outgoing. She also said bad things will continue to happen unless he acts normal.

So at her advice, he quit studying and while his grades were still quite good, they were due to his intelligence. He quit the athletic team. He became a shut in, and brainwashed that that was better. Why would Manami do this? Because she saw Kirino as competition. Young Kirino was quite normal for a girl her age, and she admired her older brother in a platonic way. That's right, Kirino's feelings didn't become romantic until after their cold war started. Manami changed Kyousuke because who he was, was what Kirino liked about him, so she changed Kyousuke so Kirino wouldn't have anything to admire. And the boring Kyousuke was what Manami wanted all along, not the outgoing and ambitious Kyousuke so it was birds with one stone.

Kirino asked Manami what happened to her brother, and Manami stated "the oniichan you admired never existed." Before this, Manami twisted Kirino's love for her brother. Kirino was far too young to know what incest was, she was just a normal girl, but Manami tricked her into thinking her admiration itself was abnormal and creepy, and that Kirino should bottle her feelings up and forget about them. Before Manami came onto the scene, Kirino couldn't keep up with Kyousuke. She tried her best to train so she could keep up. So after Manami drove Kirino and Kyousuke apart, Kirino became a model, and she continued her athletics. She became a great student, and a great runner. She was trying to remind her brother of the life he had before Manami, but unfortunately he lost confidence. The roles were reversed.

During this period, she met her Ayase and Kanako. And it was also during this period that she found out about the sexual side to romance and became obsessed with little sisters. Her first incest product was a book called "How to get your little sister to fall in love with you". She started projecting her feelings onto the imouto characters. And she got into anime like Meruru here too.

A few years later, Kirino and Kyousuke were coming out of the cold war. Their bonds had to pretty much be rebuilt. If Kirino hadn't been driven away from Kyousuke, the two would have progressed their relationship as brother and sister in a normal way, i.e. in a platonic manner. But because she was, the two missed out on psychological development, especially Kirino who became to see Kyousuke as less a brother and more a potential partner. Manami caused the very thing that she was supposedly trying to avoid. She created competition that she never had to begin with.

Then other girls step onto the scene, each falling in love with Kyousuke, and each asking Manami for advice. Her intention once again becomes known after Kuroneko enacts her plan to date and then dump Kyousuke (this was to get Kirino to admit her feelings.). After getting dumped, he calls Manami, who tells him in his emotional weakness that "any girl who dated him would surely face trouble" and that few such as herself would ever be happy marrying him. This was to lower his confidence, how could telling somebody that they're useless and that few could be happy married to him not lower your confidence if you trusted the person?

Unfortunately for her, and somewhat ironic, the reverse imprinting that was reserved for Kirino was passed onto Manami, so rather than Kyousuke seeing her as a potential romantic partner, he, in his own words, sees her more a grandmother (he asked things like "what are you my grandmother?" and he told Akagi to imagine that his grandmother turned into a fifteen year old. He basically was trying to explain the westermark effect in regards to childhood friends). Manami in a way became the "sister" when she drove Kirino away.

It is when Manami realizes that she has lost that she desperately tries to pair Kyousuke with anybody besides Kirino, because by this point the two hate each other. And Manami, feels that losing to Kirino would be worse than losing to anybody else. This is why she teaches Kanako to cook. In her mind, if she can't have Kyousuke, then neither can Kirino.

Even still, her efforts go in vain as during the rehabilitation of the Kousaka bond, the two do fall in love with each other. Kyousuke realizes he loves Kirino more than anybody else in the world. Upon hearing this Manami physically attacks Kirino and kicks Kyousuke in the stomach.

This is all explained in the novels. And there's a bit more explanation here: myanimelist.net/forum/?topicid=623227

Well...Kririno really acted bitchy in front of her, getting punched was her own fault
And even though i really love the Kousakas as a couple, the whole incest thing isn't something you should tell everyone, unlike Kirino who even brags about their relationship so...the punch was justifiable
