Ass-quaintance? That's so forced that it barely registers as a joke to me. The point of a pun is to put emphasis on a certain part of a word to give it a different meaning.

"A pleasure to be of ASSistance." would have made more sense as you wouldn't have to literally destroy pronunciation of the word to make a joke. The amateurish nature of that one line has distracted me from the glorious ass being presented! >:/

I figured it was likely a translation thing (I haven't seen the show myself so I didn't know if it was an accurate translation or not) I just wanted too be an ASShole.......................(slowly walks out).

Wow. It is funny how she is acting like a "kawaiko" when she is so large. Then again she is 5'4 in her normal form, so even tough she isn't short by Japanese standards, she isn't tall either. That said, Mount lady looks seriously cute in the anime.
