According to extra pages detailing some of the Sternritter that were released with chapter 615 in Shonen Jump, Giselle is biologically male, but the material does not confirm for certain whether Giselle personally identifies as male or female. It is also unknown if those extra pages are accurate or not since they were made by Kubo's editor instead of Kubo himself.

She calls herself a girl. So she's a girl. Also not once does she herself call herself a boy, it's Yumechika and that one foppish arrancar who do. Notably all her friends call her a girl, so that should be a good enough indicator.
