They are not blood related, he calls Kotone childhood friend in this clip and in the opening of the anime there is a big THERE IS NO BLOOD CONNECTION BETWEEN THE CHARACTERS.

The anime was way too short and hastily made imho, but had some really great animation, looking forward to the next episodes.
