Specifically, this is the Armor of Altair. To unlock it, you need to eavesdrop on the Templar meeting (aka clearing the first Assassin's Tomb), then clear the remaining five Assassin's Tombs, to get the six Assassin's Seals which are then used in the underground chamber that Mario Auditore revealed during the tour of Monteriggioni.
